Add or subtract a suitable percentage based on slope. For instance 30mph to stopped is a shorter distance if going up a 15% hill. Plus the rubber in your tyres and the temperature impact upon this etc.

27kg's what the hell are you carrying a spare bike? You can do the ride with a lot less, many do on audax rides of much greater length and time. Or take look at what people were carrying on the transcontinental or trans am. Less really is more.

Try riding slower to hit the 500km in 24 hours. You don't need to beast yourself to hit that target and beasting yourself will hit your ability to digest your food.
