Just got back from Sri Lanka -- not on the bike, but can still answer most of your questions.
S-L is a great country for the tourist. The people are really friendly, and it's easy to organise accommodation and transport.
We travelled pretty well the same route as you're planning, by bus, train and car. No prebooked accommodation necessary; there are loads of places to stay on this route, Lonely Planet guide is a start point, but don't rely on it: even the 2012 edition is hopelessly out of date. Double room prices for a start! We met a couple of touring cyclists. The road surfaces are generally excellent (better than the UK!!) : the main problem is with heat and humidity. Think 30 degrees and 90% We found (as did the cyclists we met) that by 1030-11 and until 4-430 it's painful to move around! Negombo -- stayed at Villa Shade (find it on Trip advisor). Great guest house, and they'll give you all the help you need to get started, and I'm sure they'll store your bag. the route from Kandy to Ella via hill country is a belter (make sure you've got your climbing legs on!) Mind you, after Ella you'll have 15 miles of downhill! S-L driving is interesting. we hired bikes and cycled for a couple of days without problems but keep your wits about you. Towns can be hairy. Dogs generally don't bite. More info if you want
