I agree that if the frame is a tiny bit out, you may be able to get the wheels to track properly in line by filing the dropouts.
However, two points of concern....
You still need a properly dished rear wheel to get this right.
The situation seems to change with time.

Broken Axle?

Yeah - been there, done that - a long time ago, but the symptoms are familiar................except for the broken spoke.

Thank you all for the further suggestions. To answer the points: The rear dropouts look to be OK but I don't have any means of checking accurately. The rear skewer is definitely straight and unbroken - checked that right at the start! Brake callipers appear to be making even contact on each side, I've checked the adjustment on this too. Lube on the wheel rim? I previously scrubbed this with hot water and a lot of Fairy Liquid then rinsed it off and changed the brake blocks - brand new Ultegra. To no avail.

In three weeks time I can check the bike with a spare wheel which I know is OK. I'll come back here and let you know what I find.
